What is Nrf1 and Nrf2?
Nrf1, together with Nrf2, mediates the biogenomic coordination between nuclear and mitochondrial genomes by directly regulating the expression of several nuclear-encoded ETC proteins, and indirectly regulating the three mitochondrial-encoded COX sub unit genes by activating mtTFA, mtTFB1, and mtTFB2. The Nrf proteins are also important for the upregulation of antioxidant and xenobiotic-metabolizing enzymes during oxidative stress.
NF-E2 related factor 2 (Nrf2) is a member of the cap ‘n collar family of basic leucine zipper transcription factors. By binding antioxidant response elements (AREs), Nrf2 activates the transcription of genes encoding antioxidant and phase II drug-metabolizing enzymes needed to protect cells against oxidative damage. Transcriptional targets regulated by Nrf2 include genes encoding glutathione S-transferases, superoxide diase-1, and heme oxygenase-1.
When this protein messenger called Nrf2 is activated, it enters the nucleus of every cell and it turns on several hundred of these 25,000 genes. And the genes that it turns on, or turns up, are known collectively as survival genes. What is a survival gene? These genes enable cells to survive in the face of several different kinds of stress, especially oxidative stress which is due to the over production of free radicals and other oxidants.
Nrf2 also affects another hundred or so genes by turning them down, not up. Included in these genes are pro-inflammatory and pro-fibrotic genes. Nrf2 is a protein that binds itself to a DNA sequence: Nrf2 enters cells to stimulate protective genes and enzymes to neutralize the effects of free radicals.
